Suicide bomber kills three in Iraq's Kirkuk (Reuters)

KIRKUK, Iraq (Reuters) ? A suicide bomber killed three people and wounded dozens in Iraq's Kirkuk city on Thursday after he blew up his car outside a bank where police were collecting their salaries.

The bomber drove his car into the bank premises, damaging nearby buildings and setting parked vehicles on fire in the city center, local authorities said.

Black smoke billowed from the blast site as emergency workers helped the wounded into ambulances. One body lay in the street near the wreckage from the attack.

Kirkuk, 250 km (155 miles) north of Baghdad, is ethnically divided among Iraqi Arabs, Kurds, and Turkmen and its disputed status makes it a potential flashpoint for conflict as U.S. troops prepare to withdraw at year-end.

"The security measures around the bank were tight but the suicide bomber stormed the bank by car and managed to blow himself up although security force opened fire and injured him," Hassan Turan, head of Kirkuk provincial council, told Reuters.

"The attack targeted the bank because it gives salaries to governmental offices," he said.

Three people were killed and 76 others wounded, mostly local residents, said Sideeq Omar, general director of Kirkuk health department.

Violence has fallen sharply in Iraq since the sectarian strife in 2006-2007, but Sunni Islamist insurgents tied to local al Qaeda affiliates and Shi'ite militias still carry out daily bombings and assassinations.

Attacks this year have often targeted local government offices or Iraqi security forces as insurgents have tried to destabilize Prime Minister Nuri al-Maliki's power-sharing government, split among Shi'ite, Sunni and Kurdish blocs.

Around 44,000 U.S. troops remain in Iraq more than eight years after the invasion that toppled Saddam Hussein, but they are scheduled to leave by the end of the year when a bilateral security agreement ends.

Iraqi and U.S. officials believe local armed forces can contain any internal threat now, but Baghdad and Washington are in talks about whether some U.S. troops would stay beyond 2011 as trainers to help Iraq cover gaps in its capabilities.

It was April 8, 1974, when Hank Aaron flicked his wrists and sent the ball sailing over the left-field wall.

It was something he had done hundreds of times before, but this one was special. It was the 715th home run of his career, and it had broken the most-hallowed of all baseball records, a record held by the legendary Babe Ruth, a record that had stood for 39 years.

It was a mark some thought would never be broken, having been unchallenged for so long. Before Aaron, only one player had even managed to give it a serious run, and that player ? Willie Mays ? finally ran out of gas with 660 homers in 1973.

Aaron would go on to add 40 more home runs to his record, retiring in 1976 with 755. An amazing example of consistency and longevity, Aaron had to average 33 home runs a season over his 23-year career to set the record. If not hopelessly out of reach, his record certainly seemed secure.

Then again, Babe Ruth fans probably thought the same. They couldn?t have predicted Aaron would come along.

And then again, who could have predicted Barry Bonds?

Bonds, the surly slugger whose father Bobby was a contemporary of Aaron?s, brought an incredible combination of raw power, dazzling bat speed, and a surgeon?s eye to the game. Sparked by his amazing 73-homer season in 2001 (another record), he ended Aaron?s home run reign in 2007, and finished his career with 762.

There are plenty of questions about how Bonds attained the home run record, with many people believing he used illegal performance-enhancing drugs. (In his trial this year, a jury couldn?t decide if Bonds lied to a grand jury about his drug use, though he was convicted of obstruction of justice in the case.)

But regardless of how Bonds got there, the record remains. Now the only question is will anyone ever break it?

?Seventy-three home runs will probably never be done again,? says Cincinnati Reds first baseman Joey Votto, a two-time All-Star with 118 home runs in five seasons, ?and the (career) home run record will probably never be done again, at least in the next 100 years.?

Maybe so, but as Bonds? proved, you never know for sure.

Records are made to be broken, the old saying goes. Babe Ruth found out the saying was true, and Hank Aaron predicted it would be, but is that really the case?

Let?s take a look at some of the other great achievements in baseball, and with the help of input from the players themselves, attempt to predict which marks will fall, and which will stand the test of time.

CAL RIPKEN JR.?S STREAK OF 2,632 CONSECUTIVE GAMES PLAYED

Imagine going 17 years without missing a single day of work. Now Imagine your job description includes turning double plays as base-runners try to knock you on your backside, taking countless sprints around the bases, and dodging the occasional 95-mph fastball aimed at your ribs, knee, elbow, and hands. Now you have an idea what Cal Ripken Jr., who spent most of his career at the demanding position of shortstop, had to go through to break Lou Gehrig?s record.

?In baseball now, you play so hard,? says Texas Rangers third baseman Adrian Beltre. ?You can pull a hamstring, you can sprain your wrist. There are so many things that go against even being close to that record.

?Even if you do stay healthy, you get tired. To be in the lineup every day, 162 games, is almost unheard of. You might last a couple years, but not 12 or 14 years.?

Ripken broke Gehrig?s 56-year-old record of 2,130 in 1995, then proceeded to add another whopping 501 games to his total.

To put it in perspective, only seven players have even played more than 1,000 games in a row, and the man in third place all-time ? Everett Scott ? compiled fewer than half as many as Ripken (1,307).

?It?s amazing what Cal Ripken did,? says Atlanta Braves catcher Brian McCann. ?I can?t even put into words how hard that is. To never have anything go wrong. Luck was on his side and he had the determination to go out there and perform every day. You put those things together and it?s amazing what he did.?

Verdict: Will anyone break Ripken?s record? ?I don?t see it happening,? says Beltre. We agree.

JOE DIMAGGIO?S 56-GAME HITTING STREAK/HITTING .400

We?ve included these two together because it?s been 60 years since either happened. It was 1941 when Yankees great Joe DiMaggio set his record. DiMaggio?s rival, Boston Red Sox legend Ted Williams, hit .406 that same year, and neither mark has been touched since. A Yankee and a Red Sox. Two amazing achievements that haven?t been matched in 60 years. It just seems right to keep them together.

There have been some close calls. Pete Rose compiled a 44-game hitting streak in 1978, and Paul Molitor reached 39 in 1987. In the chase for .400, Tony Gwynn hit .394 in 1994, and Rod Carew hit .388 in 1977. But that?s about it.

There are a number of reasons that will make it difficult for anyone to top either record, including an increasing reliance on specialist relievers out of the bullpen and the intense focus of attention from the modern-day 24-hour news cycle, neither of which DiMaggio or Williams had to face.

?If Ichiro can?t get it, nobody can get it,? says Votto, describing the type of player it would take to break DiMaggio?s streak. ?You can?t be a guy who walks, you can?t be a guy that strikes out. You can?t be a guy who is too deep in the order, because you?ll lose opportunities. And with the way the bullpens work now, if you?re at game 40, you?re facing some specialists-times-two, innings six-through-nine, for your last two at-bats. Good luck, you know??

Beltre agrees. ?It would take a contact hitter, a hitter kind of like Ichiro. A guy who can get a dribbler for a hit, who can get a blooper for a hit, a guy who doesn?t walk a lot, but he can get a hit any time.?

As far as hitting .400 goes, it would likely take the opposite kind of hitter, the kind who has a more discerning eye at the plate and is willing to be patient and wait for a good pitch to hit.

?It would take a lot of walks, a lot of walks, and a lot of walks,? says Cincinnati Reds second baseman Brandon Phillips. ?You gotta make sure you get at least two hits, or one hit and go 1-for-2. You gotta get two hits, or one hit and a lot of walks. Then you could do it.?

Verdict: Either mark could be reached under the right circumstances, but the planets would have to align in an almost unprecedented way. As Votto says, ?the pressure media wise and the overall attention will be exponentially more than anything Joe DiMaggio dealt with or Pete Rose dealt with. It?s too much.? That?s probably a big reason neither mark has been touched in 60 years.

Jobless claims, GDP data ease economic fears (Reuters)

WASHINGTON (Reuters) ? The chances of the economy averting a new recession got a boost on Thursday with claims for jobless benefits falling to a five-month low last week and growth a touch stronger in the second quarter than previously estimated.

Initial claims for state unemployment benefits fell 37,000 to 391,000, the Labor Department said, well below economists' expectations for 420,000. But the department cautioned that the way it adjusts the data for seasonal fluctuations may have overstated the strength.

Separately, the Commerce Department said U.S. gross domestic product grew at an annual rate of 1.3 percent in the second quarter, up from the previously reported 1.0 percent. Consumer spending and export growth both were stronger than earlier estimated.

"When you connect these data points together, they indicate a very tepid recovery. We are still experiencing positive growth, which is better than we feared a few months ago," said Paul Ballew, chief economist at Nationwide in Columbus, Ohio.

The cautious optimism generated by Thursday's data was tempered somewhat by a report showing the housing sector remained on the rocks last month.

Stocks on Wall Street were mostly higher on the data and growing confidence in Europe's response to its debt crisis. The dollar fell against a basket of currencies, while prices for U.S. government debt dropped.

FED READY TO ACT AGAIN

Political haggling in Washington over budget policy and the deepening debt crisis in Europe had eroded confidence, leaving the U.S. economy on the brink of a new recession.

A survey of U.S. chief executives released by the Business Roundtable on Thursday showed their views of the economy's prospects deteriorated in the third quarter, with the number who expect to cut jobs roughly doubling.

Faced with a weak recovery, the Federal Reserve last week announced a new measure designed to push long-term borrowing costs lower by shifting assets on its balance sheet.

In speech on Wednesday, Fed Chairman Ben Bernanke said the U.S. central bank might need to ease monetary policy further if inflation or inflation expectations fell significantly.

The drop in initial claims for unemployment benefits took them below 400,000 for the first time since early August. The department, however, said the weakness in the labor market in recent years may have led the model it uses to seasonally adjust the data to overstate last week's drop.

The decline also reflected the fading impact of Hurricane Irene, which had caused claims to spike in the September 10 week.

Still, the total number of unemployed continuing to claim benefits after an initial week of aid fell to 3.73 million in the week ended September 17 from 3.75 million a week earlier.

The September 17 week corresponds with the survey period for the department's household employment measure, which is used to construct the national unemployment rate.

RECESSION FEARS EASE

In August, the jobless rate remained stuck at 9.1 percent, with a separate survey of employers showing hiring ground to a halt, which ratcheted up recession fears.

Those worries are beginning to fade. Factory output continues to expand and businesses have maintained their appetite for spending on capital goods.

"Indications are that the third quarter is doing better than previously thought. We now anticipate third-quarter real GDP growth of just above 2 percent," said Nigel Gault, chief U.S. economist at IHS Global Insight in Lexington, Massachusetts.

Housing, however, remains a weak spot.

The National Association of Realtors said its index of pending home sales, based on contracts signed in August, fell 1.2 percent to 88.6, its lowest since April.

NAR said contract signings, which usually precede actual closings by a month or two, were held back by tight credit and, in the Northeast, by Hurricane Irene.

With millions of Americans locked into mortgages worth more than their homes, historically low interest rates are failing to lift sales. Freddie Mac said on Thursday that the average rate on U.S. 30-year fixed rate mortgages fell to a record low 4.01 percent this week.

Plane with 19 people on board crashes in Nepal

(AP) ? A plane carrying tourists to view Mount Everest crashed while attempting to land in dense fog in Nepal on Sunday, police and eyewitnesses said. A witness said 18 bodies were pulled out of the wreckage of the plane, which was carrying 19 people.

The Beechcraft-made plane belonging to Buddha Air was carrying 16 foreign tourists and three crew members and crashed in Bisankunarayan village, just a few miles (kilometers) south of the capital, Katmandu.

Poudel said it was foggy, and that visibility was very low in the mountainous area.

Rewant Kuwar, an official at the Katmandu Airport rescue office, said the plane had last made contact with the control tower at 7:31 a.m.

The plane had taken the tourists to view Mount Everest and other high peaks and was returning to Katmandu. The "mountain flight" takes tourists over the Everest region, and passengers can view some of the world's highest peaks from the airplane windows.

Global leaders struggle to calm recession fears (AP)

WASHINGTON ? The world's economic powers struggled on Friday to get on top of a European debt crisis that is threatening to dump the global economy back into recession.

Officials gathered for three days of discussion pledged to push forward to fulfill the goals of a program in which the Group of 20 major economies promised stronger cooperation to jump-start global growth and help Greece avoid a destabilizing default.

All of the discussions about European debt were occurring around the annual meetings of the 187-nation International Monetary Fund and its sister lending agency, the World Bank.

In advance of those talks, the G-20 finance officials, including Treasury Secretary Timothy Geithner and Federal Reserve Chairman Ben Bernanke, pledged a bold effort to deal with the debt crisis and encouraged Europe to move quickly to implement its promises to help Greece.

Jay Bryson, global economist at Wells Fargo Securities, contrasted the G-20 statement Thursday with the bold program the G-20 put forward in London in April 2009 at the height of that financial crisis with billions of dollars of support put forward to boost economic growth and provide a financial backstop for the IMF to rescue countries in trouble.

"You've got to back up words with actions," Bryson said of Thursday's statement, which he said was an example of "political paralysis."

"The leading lights of the eurozone are aware that time is running out," British Finance Minister George Osborne told reporters on Friday. "There is a far greater sense of urgency than there was three weeks ago about the necessity for the eurozone to address its problems."

Canadian Finance Minister Jim Flaherty said that he had stressed during the G-20 discussions that "Europe will need an exercise of political will. We need decisiveness and we need clarity."

Investors are worried that Europe's debt crisis could destabilize the global economy at a time when growth has already slowed significantly due to a jump in oil prices earlier in the year and a pronounced slowdown in the United States, the world's largest economy.

Greece could default on its debt next month unless it receives a $10.9 billion installment from other eurozone countries and the IMF.

Antonio Borges, the head of the IMF's European department, told reporters Friday that if Greece continues with the austerity measures it has pledged to put in place, then the rescue funds will continue to be provided.

"They can count on full support if they are on track," he said at an IMF briefing. "If they are not on track, then I think they will see the support evaporate very quickly."

A Greek default could destabilize other financially troubled European countries, such as Portugal, Ireland, Spain and Italy. It would also deal a blow to many European banks, which are large holders of Greek government bonds. The concern is that the shock could result in a replay of what occurred when Lehman Brothers fell in September 2009, sending waves of fear throughout the global financial system that caused credit to freeze as banks stopped lending to each other.

Geithner has said the United States has a huge stake in seeing Europe succeed. The G-20 group discussed proposals Thursday that he has raised to expand the resources of the European bailout fund by using methods the United States employed during its own financial crisis in 2008-2009.

The G-20 communique spoke of trying to increase the flexibility of the rescue fund and maximize its resources but spelled out no specific ways to accomplish those goals.

The joint statement also said the G-20 nations planned to produce a "collective and bold action plan" to boost global growth and deal with high government debt that will be put together in time for a G-20 leaders' summit in Cannes, France, on Nov. 3-4. But it provided no details of what will be in the action plan.

Singled Out

Fly over any baseball stadium when the home-team batter slams a double in the gap with two men on base, and you?ll see a crowd of fans rising in unison, arms waving wildly in the air. You?d think you were viewing typical baseball fan behavior.

Witness this scene at ground level, though, and you?ll get a different picture. While a majority of fans participate in the cheering, others are sipping beer, attending to scorecards or roaming the walkways in search of a hot dog.

Such displays of individuality shouldn?t come as a surprise ? people often react differently to the same circumstances, after all. But life?s individuality thrives at much smaller scales than the human body.

Cells, for example, are not all alike. Clusters of single cells, even of all the same type, show cell-to-cell differences in appearance, growth and behavior. This holds true for single-celled organisms too. Individuals within a colony of identical bacteria, for instance, will behave in different ways, even under the same conditions.

For decades, scientists were stuck with an aerial view of cell behavior. Experiments carried out on large pools of cells obscured information on the activities and makeup of the individuals. Difficulties in tracking molecules inside a single cell forced researchers to content themselves with averages derived from whole populations.

Now scientists are finding ways to zero in. Improvements in imaging techniques make it possible to view tiny, transparent structures and molecules within cells that are otherwise nearly invisible. New ways of separating cells during study enable scientists to sort through thousands at once, distinguishing subpopulations and studying multiple traits in just one go.

?We used to think that we could take a million cells and grind them up and make measurements on those million cells,? says Nancy Allbritton, a biomedical engineer at the University of North Carolina at Chapel Hill. ?And whatever measurements we made on those million cells would reflect what?s going on in a single cell.?

Scientists today know that averaging the constituents of a million cells often gives no clue to what?s going on at the cellular level. Cells thought to be alike have been found to produce different numbers and types of proteins. Beyond providing interesting insights into basic biology, understanding this individuality may have medical consequences ? illuminating characteristics that can make a cell more amenable to a particular drug or more vulnerable to an allergen or viral infection.

Though in many ways single-cell analysis is still in its infancy, the field is quickly expanding. Researchers are working to improve technologies and to figure out how to make sense of the data.

Lighting the way

Of the millions of different living things in the world ? large and small, plants, animals and microbes ? all have in common the fact that they are made of one or more cells. Scientists have been observing cells since the mid-17th century, when Dutch scientist Antony van Leeuwenhoek discovered bacteria and, later, blood cells. Through the years, microscopes and staining methods have allowed researchers to view and describe the basic structures and internal workings of various cell types ? understanding how skin cells, for example, differ from cells in the heart or kidneys.

Distinguishing unique features among cells of the same type has proven to be more difficult, because cells are so small. A typical human cell is about one one-hundredth of a millimeter across. Microbes are even smaller. Conventional light microscopes are constrained by a physical law demanding that the size of an object being imaged be no smaller than about half the wavelength of the light used to produce that image. To see anything as small as a molecule within a cell (the scale at which much of the work gets done), some tool other than traditional light microscopy is needed.

Scientists have found ways to get around this blind spot. When molecules and proteins are labeled with fluorescent dyes, the components inside a cell appear as bright colored dots against the background of a cell?s gel-like filling. Such advances have allowed researchers to see cellular processes unfolding at nano?meter scales. Fluorescent probes have also brought to light the wide variation that exists within a population of seemingly alike members.

When sorting through a population of cloned lung cancer cells loaded up with fluorescent probes, systems biologist Steven Altschuler noticed that each cell carried its own combination of the molecules that send signals between cells.

?It was a surprise to us,? says Altschuler, of the University of Texas Southwestern Medical Center at Dallas, who jointly runs a lab with pharmacologist Lani Wu. ?It even made us wonder what the definition of a clone was.?

The team developed a way to combine fluorescent microscopy with a type of software designed for face recognition to look for patterns among the cells. The team first labeled the cells by attaching different fluorescent tags to proteins involved in cell signaling. Computers then analyzed pictures of the cells, pixel by pixel, looking for variation in the location and intensity of the molecules. The scientists also trained the computers to identify combinations of markers that commonly appear together ? discovering the cellular equivalent of a ?blue-eyed blond.?

Using this method, the scientists were able to identify small subpopulations of cells, or stereotypes, based on which molecules appeared and where. Exposing the stereotypes to the anticancer drug Taxol revealed that different subpopulations respond differently to the drug. In May 2010 in Molecular Systems Biology, the team reported on how individual cell differences could help predict the outcome of treatment.

?That?s definitely where we would like to go,? Altschuler says. ?The dream is that you would be able to look at a small number of cells from a cancer tumor and try to predict the optimal combination of drugs to treat that particular tumor.?

Though doctors currently use various profiling techniques to determine the best treatment, Altschuler says this new approach differs because, in principle, it would allow a doctor to look at every cell in a tumor biopsy, identifying clues that would get lost in a ?population averaging? approach.

Multiplying traits

But fluorescent microscopy has its own limitations. Scientists can look for only a handful of variations, or biomarkers, in a cell at one time. Though cells carrying biomarker A could be distinguished from those carrying B or C, for example, scientists can?t simultaneously scan the cells for a second characteristic that might be important in understanding the first.

Altschuler and Wu?s team has found ways to maximize the use of fluorescent probes to get more details on cell-to-cell differences. Using three out of four available probes to home in on subpopulations of interest, a fourth probe can be free to search for a second variation within each of those groups. By running a series of experiments, rotating the fourth marker?s target each time, scientists can get more detailed information on members in each group.

Say you?re trying to understand people at a baseball stadium, instead of cells, and you want to figure out differences in the way the players, vendors and fans dress. Three out of four fluorescent probes would be used to separate people into each of these groups. The fourth probe could then search for a specific item, such as sneakers, which may be worn by members in each group. In subsequent experiments, the first three probes remain set to identify players, vendors and fans, while the fourth spots people sporting home-team jerseys, then baseball caps and so on. A computer program stitches together the information gleaned from the serial studies to show what people in each group wear.

In this process, a scientist will never look at the jerseys and caps on any given person at the game, because only one measurement can be made at once. ?Still, reliable information can be gained,? Altschuler says. His group is now profiling cells in this way.

At her University of North Carolina lab, Allbritton and colleagues are developing ways to study multiple variables beyond the limitations of microscopy. By employing what they call ?separation techniques,? the researchers have the potential to measure the activity of many enzymes ? a dozen or so ? at once. Each cell gets its own well in a grid containing thousands of rows and columns.

Cells are first loaded with various enzyme substrates, substances that work with an enzyme to produce a specific reaction. The cells are then killed and their contents released. By separating out each of the chemicals that were put into the cells as well as the products the cells make, scientists get a readout on what?s going on inside.

Because each cell sits in its own well, scientists can analyze the cells separately. Comparing the reactions of different cells is key, Allbritton says, because a study of a single cell won?t reveal much variability.

?That?s like looking at one human and trying to understand how variable people are,? she says. ?You would assume if you got a redheaded person that everyone had red hair ? and was 5-foot-4 and was female.?

Allbritton is using this technique to measure protein activity in human cells. Her studies currently focus on kinases, proteins that play a vital role in cell signaling and the regulation of gene activity.

Live on and inform

Such clues to what?s going on inside single cells often come at a price: Cells are damaged or killed in the process, providing only a single snapshot in time. This makes it hard to predict how a single cell will respond to a given stimulus.

J. Christopher Love, a chemical engineer-turned immunologist at MIT, combines the idea of separating cells and the helpfulness of fluorescent dyes to follow biochemical processes over time in living cells. His lab takes an array of subnanoliter-sized wells and fills each with either a single immune cell or a small group of them. With tens of thousands of wells, the unit looks like an ice cube tray the size of a microscope slide. Fluorescent markers identify the various proteins in or on each cell, distinguishing one cell type from another.

Over the course of several hours or days, Love studies the proteins and chemicals secreted by the cells to see how they change. Borrowing a technique from the printing of U.S. dollar bills, he presses the microarray against a glass plate to get a printout (so to speak) of the proteins and chemicals secreted by the cells. Unlike Allbritton?s work, the method doesn?t kill the cells because information is captured from the outside, on the glass plate.

Love is applying this interrogation method to ask questions that are relevant to chronic diseases such as HIV, autoimmunity, cancer and allergies. One ongoing study tracks how allergic patients become desensitized to milk allergens, measuring how the patients? white blood cells respond to milk extracts. The study looks at how many cells are responding, what kinds of immune proteins are being produced and how much of each protein is produced by each cell.

By analyzing large numbers of single events and interactions between small groups of cells in concert, Love can spot rare types of cells and events that are linked to disease. Certain T cells and B cells found in diseases like multiple sclerosis or diabetes, for example, typically are present at rates of 1 in 10,000 to 1 in 100,000 of the circulating T cells in a blood sample, he says. Without looking at many, many cells, he wouldn?t be able to spot or study the interactions of these rare types.

?Single-cell analysis is a bit of a misnomer in the sense that you?re not really looking at one cell,? Love says. ?You?re resolving to one cell, but you are needing to look at thousands, if not tens or hundreds of thousands, of cells to understand what that individual measurement looks like in relation to the rest of the distribution.?

Single microbes

Studies of cell-to-cell differences also promise to reveal new information about single-celled organisms. For a long time, biologists thought that members of a microbe colony were all basically the same, and that those cloned from a single cell, because they were genetically identical, should react to environmental challenges in the very same way.

Scientists now know that individual traits may, in fact, serve as drivers to boost the robustness and resistance of microbial populations, says Michael Konopka, a biochemical engineer at the University of Washington in Seattle. In the October 2010 issue of Nature Chemical Biology, he and coauthor Mary Lidstrom outlined how some members of a microbial colony will stop growing in times of stress, moving into a dormant state that allows them to ride out the bad conditions while their neighbors perish.

Understanding how individuals adapt to environmental conditions, such as temperature fluctuations, could help engineers find ways to boost certain types of chemical processes used in manufacturing, Konopka says. Some naturally occurring microbes, for example, may have a built-in capacity to do the chemical reactions that industrial researchers want to perform.

Others research teams are developing ways to determine the complete genetic blueprint of single-celled organisms to get insight into whole populations of cells that are otherwise difficult to study. Stanford University scientists working under the direction of Stephen Quake are plucking single cells from populations found in pond scum, soil or even plaque on teeth, to ask a seemingly simple question: Who are you?

Conventional genetic analysis techniques require a large, pure sample of the microbes, says Paul Blainey, a postdoctoral researcher in Quake?s lab. ?That usually requires that you?re able to isolate the particular organism and grow up a whole huge beaker of it in the lab.? Unfortunately, the chemical reaction used to amplify tiny amounts of DNA is notoriously prone to contamination.

To get around this problem, the Stanford group is using a laser tweezer to sort individual microbes inside tiny, automated devices designed to analyze minute traces of DNA. Enclosed within this miniature lab, microbes and their DNA are safe from contaminants.

In 2007, Quake?s group analyzed the genome of a single bacterium found in dental plaque taken from a person?s mouth. This year the team turned to a less familiar class of single-celled organisms, the Archaea: Online February 22 in PLoS One, the researchers published findings on the genome of a single-celled ammonia-eater from San Francisco Bay.

Genetic information gleaned from an individual microbe, combined with findings on cell-to-cell differences in behavior, may provide new insights into the thousands of unknown or barely known populations found in water and soil. Such insight could in turn offer clues to how microbes act as infectious agents and how they develop resistance to antibiotics.

Despite recent gains in studying single cells and how they differ, there?s much work to be done, Allbritton says. Current techniques still allow only a small number ? fewer than two dozen ? of the thousands of molecular components that float around in cells to be measured at one time.

?It?s embarrassing,? she says. ?We?ve made a lot of progress, but you can see how far we have to go.?

With such a small subset of cellular components under study, scientists? perception of how cells operate may be colored by what they see, she adds. The big push now is to develop ways to measure more of the cell?s contents, and see how they change over time and work in relationship to each other.

?If we can see everything,? Allbritton says, ?it might dramatically change how we view the single cell.?
